SOLIAN (AMISULPRIDE): DRUG INTERACTIONS
A number of medications can increase the risk of ventricular arrhythmias including torsades de pointes.
The use of the following pharmaceutical drugs is contraindicated:
Medicines which could induce torsades de pointes:
- Class Ia antiarrhythmic agents such as disopyramide and quinidine.
- Class III antiarrhythmic agents such as sotalol and amiodarone.
- Other medications such as bepridil, sultopride, cisapride, thioridazine, erythromycin intravenous, methadone, vincamine intravenous, halofantrine, sparfloxacin, pentamidine.
Levodopa: reciprocal antagonism of effects between levodopa and neuroleptics
Caution is required with the use of the following medications:
Medicines which enhance the risk of torsades de pointes or could prolong the QT interval:
- Medicines which induce bradycardia, such as bradycardia-inducing calcium channel blockers (verapamil, diltiazem), beta-blockers, guanfacine, clonidine, digitalis.
Pharmaceutical drugs which can cause hypokalaemia, such as stimulant laxatives, diuretics, amphotericin B intravenous, tetracosactides, glucocorticoids. Hypokalaemia should be corrected.
Neuroleptics such as chlorpromazine, thioridazine, trifluperazine, haloperidol, pimozide, imipramine lithium salts, antidepressants.
Concomitant use of Amisulpride (Solian) with other anti-psychotics may increase the risk of developing neuroleptic malignant syndrome.
Amisulpride may enhance the effects of alcohol.
Solian (Amisulpride) tablets and oral solution may enhance the effects of the following medicines:
CNS depressants including narcotics, analgesics, anaesthetics, sedative H1-antihistamines, benzodiazepines, barbiturates, and other anxiolytic medicines, clonidine and derivatives.
Antihypertensive medications and other hypotensive medicines.
A placebo-controlled study of concomitant use of lithium carbonate 500 mg twice daily and a low dose of amisulpride (100 mg) twice daily in healthy young male volunteers showed no effect of amisulpride on the pharmacokinetics of lithium. A small trend towards prolongation of the QTc interval was observed when lithium and amisulpride were co-administered but is not regarded as clinically important.
A study of the effect of concomitant use of cimetidine on Solian (Amisulpride) oral solution and tablets excretion has not been conducted.
In vitro studies using human liver microsomes and cryopreserved human hepatocytes did not show evidence of significant amisulpride metabolism. Based on these results, it is unlikely that drug interactions involving amisulpride would occur due to inhibition or induction of cytochrome P450 - mediated metabolism.
